Abstract

Lichen planus (LP) is a chronic inflammatory mucocutaneous disease of unknown etiology. It is estimated that LP affects approximately 1% of world population and may involve skin, nails, scalp and mucous membranes separately or simultaneously. There are several immunological parameters related to the innate or adaptive immune response in the LP, which would be essential to better understanding of the immunopathogenesis of the disease. The proposal of this project is to evaluate in the Lichen planus the influence of the innate immune response by myeloid (mDC) and plasmacytoid dendritic cells (pDC) and their influence in the generation of pro-inflammatory cells, regulatory cells as well as the cytotoxic effector cells. To evaluate the mononuclear cells response to the agonists of TLR 2-9 and in the inflamassoma generation in the cytokines production (IFN-alpha, TNF-alpha, IL-1beta); analyze the effect of these signaling pathways in the mDC and pDC, as well as evaluate the frequency of DCs in blood and tissue lesions. Furthermore, to assess the impact of DCs differentiated from monocytes or purified pDC in the activation of allogeneic cells. The CD4+ T cells response will be evaluated by the polyfunctionality to secreting cytokines (IL-17, TNF-alpha, IFN-gamma, IL-21) and the regulatory CD4+CD25+CD45RA-CD127-Foxp3+ T cells. The cytotoxic response mediated by NK cells and CD8 + T lymphocytes, will be analyzed by the phenotypic and polyfunctional profile. Furthermore, the purified CD8+ T cells will be assessed for the expression of cytokines/chemokines and receptors by the method of PCR array. (AU)
