Analysis of inflammasomes activation and regulation and effects on dendritic cells in the induction of Th17 cells in lichen planus

Abstract

Lichen Planus (LP) is a mucocutaneous disease of chronic inflammatory nature and unknown etiology. Although the high incidence of this disease, there is scarce immunological investigations, mainly concerning the influence of components of the innate immune system in the inflammatory process and its influence on the adaptive response. Furthermore, the products generated by the activation of these pathways may influence the maturation of dendritic cells as well as the cells with pro-inflammatory or regulatory potential. The aim of this project is to analyze in the LP the activation of the inflamassoma and establish a strategy to regulate the inflammassoma generation through the TLR7/8 pathway. Moreover, to evaluate the proinflammatory and regulatory potential of the TCD4+ cells in the LP. Inflamassoma activation induced by lipopolysaccharide and adenosine triphosphate and its regulatory effect exerted by TLR7/8 (CL097) will be initially evaluated in peripheral blood mononuclear cells (PBMCs) by the IL-1² secretion and NALP3 and Caspase 1 expression. The effect of these pathways in monocyte-derived dendritic cells (moDCs) will be evaluated by the expression of costimulatory molecules and cytokine secretion as well as by the ability to heterologous Th17 generation. In addition, the frequency of dendritic cells in peripheral blood and the Th17 cells/regulatory CD4+ CD25+ Foxp3+ T cells will be assessed in patients with LP. (AU)
