Expression of matrix metalloproteinases and PCNA in deep corneal ulcers, induced in rabbits, treated with platelet-rich plasma


The cornea is the anterior transparent structure of the eye and it's susceptible to traumas and aggressions, that can cause corneal ulcer. Its repair, after lesion, involves a series of complex and coordinated cellular processes that include proteinases and their inhibitors, growth factors and cytokines. The main proteinases presents in the cornea and in the process of its repair are the serine and matrix metalloproteinases (MMPs), which have their actions influenced by several growth factors. The platelet-rich plasma (PRP) contains growth factors that are actively secreted by platelets to begin the healing process, accelerating the epithelialisation. The aim of this study is to evaluate the influence of platelet-rich and poor plasma in the celullar proliferation, as well as the expression of matrix metalloproteinases in the cornea during corneal repair after induction of deep corneal ulcers in rabbits. Will be used 45 females of the leporine specie, constituting three experimental groups of 15 animals each one, designated Platelet-Rich Plasma Group (GR), Platelet-Poor Plasma Group (GP) and Control Group (GC). In GR the rabbits will be treated with eyedrops of autologous platelet-rich plasma, while in the GP will be used eyedrops of autologous platelet-poor plasma. In GC, lubricant eyedrops will be instilled. The experimental groups will be divided into three subgroups of five animals each (M4, M7 and M30), according to the final evaluation periods. After euthanasia and enucleation, the corneas will be processed for the immunohistochemical analysis of PCNA expression and to the MMPs: collagenase (MMP-1), gelatinase A (MMP-2), gelatinase B (MMP-9) and MT1-MMP (MMP-14) and for a tissue inhibitor of matrix metalloproteinase (TIMP-1).
