Pro-inflammatory and inflammatory cytokines, and intestinal microbiota in the acute phase of experimental colitis treated with probiotic

Abstract

The experimental colitis induced by 2,4,6 - trinitrobenzenosulfônico acid (TNBS) has the advantage of producing lesions similar to inflammatory bowel disease in humans and reproducibility. The imbalance of pro and anti-inflammatory released by the inflamed intestinal mucosa will determine the intensity and duration of the inflammatory response. Probiotics act in a beneficial immune response with increased production of inflammatory cytokines and reduced production of proinflammatory cytokines in the intestine. This study aims: to study the levels of pro and anti-inflammatory, modulation of intestinal microflora and the activity of myeloperoxidase (MPO) in acute model of experimental colitis induced by TNBS, with therapeutic and prophylactic treatment with two probiotics. The study will include 100 male Wistar rats were divided into five groups: 20 + S.boulardii mesalazine group, 20 in group L. reuteri + mesalazine, 20 in the group without probiotic, shan 20 animals in the group and 20 in group + L. S.boulardii reuteri + mesalazine. Induction of colitis will be performed by rectal administration of 50 mg / kg TNBS (Sigma ®) associated with 50% ethanol. The animals receive probiotics seven days before induction of colitis. After induction the animals continued to receive probiotics associated with mesalazine, the sacrifice of animals will be divided into two parts, half the animals will be sacrificed on the third day after induction of colitis and the rest on the seventh day after induction of colitis. The materials will be collected for analysis on the day of sacrifice of animals, according to the methodology. (AU)
