Abstract

The objectives of this study are to develop methodologies for extraction and quantification of metabolites in LC/MS/MS to determine the metabolic profile of sugarcane and eucalyptus plants treated or not treated with the herbicide glyphosate and to evaluate the effects of herbicide compounds important for plant defense and metabolism of glyphosate. The analytical methods are developed in a high performance liquid chromatograph coupled to a triple quadrupole mass spectrometer (Q TRAP 3200 LC/MS/MS). The eucalyptus and sugar cane will receive the application of glyphosate at doses of 7.2, 18, 36, 72, 720 and 1440 g a.e. ha-1, and will be kept plants without application of the herbicide (controls). At 2, 7, 14 and 21 days after glyphosate application will be collected four plants (repetitions) of each treatment for determination of the following compounds: quinic acid, shikimic acid, shikimate-3-phosphate, corísmic acid, transcinâmic acid, Á-coumaric acid, caffeic acid, ferulic acid, chlorogenic acid, salicylic acid, protocatechuic acid, gallic acid, amino methyl phosphonic acid (AMPA), glyphosate, and the three aromatic amino acids (phenylalanine, tyrosine and tryptophan). There will be a trial to assess plants of different sugarcane varieties and eucalyptus clones can metabolize glyphosate, promoting root uptake of the herbicide in order to prevent the glyphosate residue on the outside of the leaves is metabolized by microorganisms and AMPA from this degradation is absorbed and found in the leaves, which has been identified in previous tests. The data will be submitted to correlation analysis between the metabolites, analysis of variance by F test (5% probability) and means were compared by t test (5% probability). (AU)
