Visible light during growth enhances conidial tolerance to different stress conditions in fungi

Abstract

Visible light exposure during growth influences primary and secondary metabolism, growth, sexual and asexual development, and pigment formation in many fungal species. However, little is known about the phenotypic effects of light during mycelial growth on the tolerance of the developing fungal conidia to different stress conditions. In this study, conidia of several fungal species from two different phyla (Ascomycota and Zygomycota) will be produced on potato dextrose agar plus yeast extract (PDAY) medium under continuous, alternated exposure to visible light or dark. The effect of different light wavelengths, from blue to red, will also be studied. The tolerances to UV-B radiation, heat, oxidative stress, osmotic stress, and genotoxic agent 4NQO of the conidia produced under the different light exposure conditions will be evaluated and compared with the tolerances of conidia produced on minimal medium in the dark, which is known to produce conidia tolerant to a variety of environmental stresses. The cellular mechanism in which light changes the stress tolerance will be studied based on differential gene expression. The results in the fungal stress-tolerance phenotypes and in the gene expression could provide information about the effect of light during mycelial growth on the stress tolerance of the developing conidia of plant pathogens, fungal antagonists, insect pathogens, and saprophytic fungi. The main hypothesis is that light during growth improves conidial tolerance to one or several stress conditions in some fungi. (AU)
