Abstract
AGO's which are derived from partially N-acetylated poly-glucosamine (i.e. chitosan) have attracted much interest to their biological activities, mainly being involved in signaling pathways, such as morphogenetic action in embryo development in animals and regulation of the synthesis of extracellular matrix in chondrocytes. An AGO hexamer of the composition (GlcNAc)3(GlcN)3 can occur principally in form of 20 isomers. Neither the sequence analysis by NMR spectroscopy, nor chemical synthesis of higher AGO's, such as hexamers, has not yet been achieved. Our aim is to analyze AGO's which are isolated from hydrolysates of chitosan by most advanced mass spectrometry. Besides the elucidation of sequences of the individual isomeric components present in complex mixtures, we will investigate the limits of the methodology with respect to the length of an oligosaccharide and the complexity of the mixture.
