Fibroblast growth factors (FGFs) in the differentiation of the bovine cumulus-oocyte complex

Abstract

We have recently demonstrated that oocyte derived FGFs stimulate cumulus expansion and alter the expression of regulatory genes. This proposal aims to assess the effects of FGFs 2, 10 and 17 and BMP15, as well as of the interactions FGFs/BMP15, on nuclear maturation, cumulus expansion, energetic metabolism, apoptosis, steroid production and expression of markers of developmental competence in bovine COCs submitted to IVM. Studies focusing the physiological roles of these FGFs and BMP15 are also proposed. These studies will test the effects of FGFs 2, 10 and 17 and BMP15, as well as of the interactions FGFs/BMP15, on the same parameters listed above (excluding nuclear maturation) on oocectomized COCs. The removal of the oocyte aims to neutralize the effects of oocyte factors secreted during culture that would mask the effects of the treatments. Moreover, we propose to assess the expression of genes known to regulate the functional parameters altered by the treatments. Finally, to investigate the paracrine action of these FGFs in the control of cumulus cells differentiation, the hypothesis that the bovine COC secrete these FGFs in culture and that the secretion is regulated by FSH will be tested.
