Deslorelin acetate effect on domestic CAT embryo production in vivo and in vitro

Abstract

We aim to performe a gestational evaluation and produce in vitro embryos from domestic cats previously treated with deslorelin acetate. There will be performed two experiments. At experiment 1 will be used 5 domestic queens. These females will remain in quarentine and in this period the sexual behavior and vaginal cytology (VC) will be performed every 3 days. Progesterone will be measure trough feces every 7 days. When in interestrus/diestrus the queens Will receive the contraceptive treatment with deslorelin (4.7mg/animal). Queens will be monitored trough VC, sexual behavior and hormonal assays weekly during at least 180days. After this period when showing estrus characteristics the queens will be placed together will a fertile tom cat so they can copulate. After this, the queens will have their progesterone monitored every 48 hours. Pregnancy will be monitored trough ultrasound exams. If any queen present signs of abortion and if necessary a caesarean and a ovariohysterectomy (OSH) will be performed. In these cases the uterus and ovaries will be evaluated histologically and for hormonal receptor imonnohistologically. Microbiological exams will also be performed to confirm possible abortion due to infectious causes. At experiment 2 20 queens placed in two different groups: treated (A) and control (B) and 3 tom cats for semen collection will be used. During the quarentine the sexual behavior, vaginal cytology, progesterone assays and contraceptive treatment will be performed in the group A (n=10) as described in experiment 1. However after 180 days of treatment the queens Will be submited to OSH to subsequent oocyte recuperation, MIV and FIV. After the in vitro embryo production a sample of bastocysts will be evaluated trough tunel technique. In group B the queens (n=10) will be evaluated the same way described previously, but the females will not be treated with deslorelin acetate before the OSH. The blastocysts will be evaluated the same way has in group A. Data will be tabulated and the appropriated statistical evaluation will be performed. (AU)
