Cardiovascular regulation under physiological pathophysiological conditions

Abstract

The present study was grouped into five projects. In projects 1 it will be investigated the post excitatory depression (PED) of the aortic depressor nerve (ADN) after electrical stimulation of the baroreceptors afferents. In this project it will be examined, particularly, the role played by autocrine/paracrine factors (PGI2 e ROS) in the development of PED. In projects 2 a technique for immunohistochemical staining for substance P in the ADN of normotensive rats will be developed and standardized, followed by the quantification of the amount of substance P in myelinated fibers. The techniques for staining substance P will be standardized for the exam of unmyelinated fibers by means of transmission electronic microscopy. In projects 3 it will be examined the hemodynamic responses to electrical stimulation of the aortic depressor nerve of conscious L-NAME hypertensive rats, submitted to the pharmacological blockade with methyl atropine, atenolol and prazosin. These protocols aim to evaluate the relative role played by autonomic afferences (sympathetic and parasympathetic) in arterial pressure and heart rate regulation in the L-NAME hypertensive model. In addition, it will be evaluated the responses of mesenteric and hindquarter resistance in the control of blood flow during the electrical stimulation of the ADN, by means of the Doppler flow meter. In projects 4 it will evaluated in conscious rats, during the development (2, 7 and 14 days) of L-NAME hypertension, the renal sympathetic nerve activity by means of telemetry. Finally, in projects 5 it will be investigated in rats and mice with chronic heart failure developed after coronary artery ligation, the chronic effect of acetylcholinesterase inhibition with pyridostigmine on autonomic balance, heart rate and arterial pressure variability in time (variance) and frequency domain (spectral analysis). In addition, it will be also investigated the reflex control of heart rate by means of the evaluation of spontaneous sensitivity of the baroreflex using the sequence method. The studies of cardiovascular variability in mice will be undertaken using telemetry. Still in projects 5 it will be evaluated in anesthetized rats and mice the influence of chronic acetylcholinesterase inhibition with pyridostigmine upon the cardiac function by means of the pressure vs. volume curve of the left ventricle.
