Cytotoxicity of peracetic acid: evaluation of metabolism, structure, cell death and cell stress in L929 fibroblasts

Abstract

Recently peracetic acid (PA) has been reported in the dental literature as a possible root canal irrigating solution. It has the advantage of bringing together in one product antibacterial activity and ability to remove smear layer. For the selection of root canal irrigating solution it should take into account not only its therapeutic benefits, but also the possible cytotoxic effects, since it can contact the periapical tissues. The aims of this study are: 1) To evaluate the cytotoxicity of the PA, at different concentrations, in comparison with sodium hypochlorite (NaOCl) through study of the metabolism and cell death in L929 fibroblasts; 2) To investigate the mechanism of cytotoxicity of AP in L929 fibroblasts through the evaluation of metabolism, structure, ultrastructure, cell death (necrosis and /or apoptosis) and stress of these cells. L929 fibroblasts in culture will be exposed to different concentrations of PA and NaOCl and to the culture medium for 24 hours (control). After this period, it will be evaluated the cell metabolism by means of metiltetrazólio assay (MTT), the external morphology of cells by scanning electron microscope, the ultrastructure by transmission electron microscope, the cytoskeleton by staining for actin and ±-tubulin, the type of cell death (necrosis or apoptosis) by flow cytometry and the cellular stress by marking of heat shock protein 70 (Hsp70). The statistical analysis will be performed using a significance level of 5%. Statistical tests will be chosen according to the distribution and homoscedasticity of the data. (AU)
