Abstract

The diagnosis of Alzheimer's disease (AD), a great public health problem, is imprecise and delayed (since the neurodegenerative processes begin years before the appearance of symptoms). The validation of AD biomarkers has the potential to afford early diagnosis and novel treatments. This project aims to investigate, in a multidisciplinary and translational fashion, the accumulation of b-amyloid peptide (the pathological hallmark of AD) and its associated brain changes (macroscopic and molecular), bringing together several 1A-B CNPq scientists and other researchers, members of 11 post-graduate teaching areas (7 of which level 6-7 CAPES), distributed across four centers of the Universidade de São Paulo - Faculdade de Medicina da USP (FMUSP), Faculdade de Medicina de Ribeirão Preto (FMRP), Instituto de Ciências Biomédicas (ICB) and Instituto de Psicologia (IP). We will carry out clinical studies with large samples, and these will be integrated with basic neuroscience investigations aimed at detailing the molecular processes underlying biomarker changes associated with the progression of AD and its treatment. These studies will involve: the use, for the first time in Brazil, of the b-amyloid tracer 11 C-PIB with positron emission tomography (PET); studies on healthy and pathological brain aging integrating the FMUSP-FMRP groups and evaluating 500 elderly individuals (subjects with minor cognitive impairment, AD and healthy controls) using magnetic resonance imaging and other biomarkers (cerebrospinal fluid / blood); application of novel pattern recognition methods for the extraction of statistical data evaluating the relative value of different biomarkers in regard to diagnostic accuracy, thus helping to delineate individualized "neurobiological signatures", within a perspective of personalized Medicine; and experiments with animal models and cell cultures investigating the impact of the exposure to b-amyloid in the brain, as well as changes in associated biochemical and electrophysiological processes. By endorsing the interaction of this group of scientists in the short and long term, we aim to increase the impact of our publications, pave the way for innovative drug trials with disease-modifying agents, and promote technological innovation in terms of diagnostic processes and patenting of novel pharmacological agents.
