Development of selective phases for in-tube SPME/LC-MS/MS analysis of parabens and drugs in biological samples

Abstract

In recent years, modern analytical chemistry has seeked simplification by hyphenation of the techniques, automation (high-throughput performance), and miniaturization of the analytical systems. In this context, the in-tube solid phase microextration (in-tube SPME) hyphenated to liquid chromatography coupled to mass spectrometry, tandem system (LC-MS/MS, reference analytical technique for bioanalysis) can be highlighted. The development of stationary phases for in-tube SPME, such as the restricted access materials (RAM), the molecularly imprinted polymers (MIP), and hybrid silica monolith have enabled direct biological fluid introduction into chromatographic systems and selective pre-concentration of the analytes, respectively. In this project, the phases MIP-RAM and monolithic hybrid silica containing cyano and phenyl functional groups will be synthesized for in-tube SPME/LC-MS/MS analyses of parabens in breast milk samples and of drugs in plasma samples from schizophrenic patients, respectively. The proposed objectives emerged from two distinct public health problems. The first one concerns to research carried out in vitro and in vivo, which have demonstrated that parabens, a chemical preservative present in cosmetic products, can mimic the action of estrogen and induce the growth of MCF7 cells, which are related to the development of breast cancer. Further than this fact, in lactating women, breast milk may be the major route for the excretion of bioaculative lipophilic substances (such as parabens). The second problem concerns to schizophrenia, considered as the most serious of the neuropsychiatric upheavals. Apart from antipsychotics, great part of the schizophrenic patients also makes concomitant use of other classes of drugs, such as antidepressants, anticonvulsants, and anxiolytics, in order to decrease the symptoms associated with this condition. Consequently, the analysis of these drugs in plasma samples from schizophrenic patients is important to adjust the doses, minimize adverse effects and check the patient compliance with treatment.
