Abstract
In recent years, modern analytical chemistry has seeked simplification by hyphenation of the techniques, automation (high-throughput performance), and miniaturization of the analytical systems. In this context, column switching liquid chromatography coupled to tandem mass spectrometry (column switching LC-MS/MS) can be highlighted. The development of stationary phases for the first dimension (1D) of this analytical system, such as the restricted access materials (RAM), the molecularly imprinted polymers (MIP), and the polymeric ionic liquids (PIL) have enabled direct biological fluid introduction into chromatographic systems and selective pre-concentration of the analytes. In this project (i) MIP-RAM and PIL phases (1D) were synthesized and evaluated for column switching LC-MS/MS analysis of the endocannabinoids in plasma samples from patients with Parkinson's disease (PD) under therapy with cannabidiol, and (ii) RAM phase (1D) and reverse-phase with core-shell particles (2D) were evaluated the for column switching LC-MS/MS analysis of the drugs (antipsychotics, antidepressants, anticonvulsants, and anxiolytics) in plasma samples from schizophrenic patients. The proposed objectives emerged from two distinct public health problems. The first one concern to evaluate new drugs, such as cannabidiol, to treat PD, since the treatment with dopaminergic drugs, especially levodopa, markedly improves the motor symptoms, but does not halt disease progression. The second problem concerns to schizophrenia, considered as the most serious of the neuropsychiatric upheavals. Among antipsychotics, great part of the schizophrenic patients also makes concomitant use of other classes of drugs, such as antidepressants, anticonvulsants, and anxiolytics, in order to decrease the symptoms associated with this condition. Consequently, the analysis of these drugs in plasma samples from schizophrenic patients is important to adjust the doses, minimize adverse effects and check the patient compliance with treatment.
