Gene expression of MDR-1, TP53, Bcl-2 and Bax in canine transmissible venereal tumor and its relationship to aggression and response to therapy

Abstract

The transmissible venereal tumor - TVT along the years has been subject of numerous investigations, nevertheless, there are gaps that need more studies, there are TVTs more malignant, do not respond satisfactorily to conventional therapeutic, and it seem that when these cells exhibit these characteristics, are present and so exacerbated progressive modifications of the biological profile. With the advent of research, such as cell culture, flow cytometry and molecular biology, is possible to identify and quantify these modifications, even predict the biological behavior of this entity. The aim of the study is to quantify the expression of genes BAX, BCL2, TP53 and MDR1 in TVT cells (before culture), and in primary culture of TVT for 3 passages before and after therapy with vincristine that help to identify modifications related with to resistance and tumor malignancy. For both will be used 8 tumors coming of dog, males and females with cytological diagnosis of TVT, seeking to evaluate tests of cytotoxicity, cell survival, growth curve, apoptosis and expression of genes by RT-PCR. It is expected that the results may contribute to understanding the dynamics of tumor growth and its behavior in anticancer treatment. (AU)
