Characterization of apoptosis related genes in canine cutaneous mast cell tumors and its value as prognostic indicators

Abstract

Mast cell tumors are potentially malignant neoplasms which have variable macroscopic features and biological behavior. These factors have turned very difficult the formulation of accurate prognosis and treatment. Alterations of apoptosis favor tumor progression by allowing the survival of the neoplastic cells in the circulation and other microenvironments. The pro-apoptotic protein BAX forms nonselective pores in the outer mitochondrial membrane. This phenomenon initiates the release of cytochrome C into the cytoplasm and activates Apaf-1 and the caspases 3 and 9, resulting in cell death. On the other hand, BCL2, an anti-apoptotic protein, antagonizes the effects of BAX and prolongs the life of the cell. The objective of this study is to detect and quantify the protein expression of BAX, BCL2, Apaf-1, caspase 3 and caspase 9, as well as their respective m-RNA in canine cutaneous mast cell tumors and to correlate these with the histopathological grading, clinical evolution and mortality. Thus, one hundred cases of canine cutaneous mast cell tumors will be submitted to histological, immunohistochemical and molecular analyzes. (AU)
