Implantation of three-stage compound continuous culture and study of probiotic, prebiotic and simbiotic action on colon cancer

Abstract

Colorectal cancer is one of the most common fatal malignancies in result of neoplasm in occidental countries, being that in Brazil it occupies the third position between the causes of deaths. Some studies has been demonstrated that the diet can influence in the cancer risk, in special the consumption of probiotic, prebiotic and simbiotic, which can influenced in the risk of the cancer developed. The objective of this project is to implant one in vitro system that it will simulate the intestinal conditions and to use this system in the selection of the probiotic, prebiotic and simbiotic, as well as its validation using animal model and to test the action of these in the diverse stages on the colon cancer development. This project will be carried through in two phases, Phase I: in vitro study, Phase II: study using animal model. In Phase I a Three-Stage Compound Continuous Culture System will be mounted and in this system they will be selected probiotic, prebiotic and simbiotic. In vitro phase will be monitored the bacterial populations by microbial plate count and the Short-chain fatty acid production by high-performance liquid chromatography (HPLC). In phase II the prebiotic and the simbiotic combination that to get the best resulted in Phase I, they will be evaluated in colon cancer chemically induced animal model. This phase will go determine in which stage of carcinogenic (initiation, promotion or progression) the consumption of probiotic, prebiotic or simbiotic can exert anti-carcinogenic effect. The checking of the carcinogenic development will be carried through by following evaluations: Aberrant Crypt Foci (ACF) assay, comet assay, histological analyses and measures of the tumor (weight, width and volume). Ali the results will be analyzed by the Analysis of variance (ANOVA) and by tests Tukey's averages. (AU)
