Evaluation of the effects of the cytokine SOFAT on osteoblast cells and its potential on bone inflammatory diseases

Abstract

A novel activated human T cell-secreted cytokine, referred as secreted osteoclastogenic factor of activated T cells (SOFAT), that induce osteoclastogenesis in a RANKL-independent manner was recently described. We previously demonstrated that the mRNA and protein levels of SOFAT were significantly higher in the gingival tissue of the periodontitis group when compared to non-periodontitis one. In addition, SOFAT potently induced TRAP-positive multinucleated cell formation by RAW 264.7 cells as well as induced the formation of osteoclast-like cells in the periodontal ligament in mice. The present study aims to evaluate the role of this new cytokine on bone inflammatory diseases. Thus, it will be exploited the activity of this cytokine on osteoblast cells in order to evaluate if this cytokine may induce higher levels of pro-inflammatory cytokines as well as to evaluate the RANKL expression in the presence of SOFAT. Besides, it will be evaluated which sub-type of T lymphocyte is able to produce SOFAT. (AU)
