Abstract
Antibodies (immunoglobulins) are an important class of biomolecules employed in the medical, clinical diagnosis, therapy, biosensors, and others applications. Annual sales of antibody therapeutics already exceed US$ 50 billion worldwide, accounting for >6% of pharmaceutical revenues. Since the purification processes available for these molecules are, in general, multistep, long, high-cost, and low yield, there is considerable need to develop new purification methodologies, such as Mixed-Mode Chromatography (MMC). MMC is a chromatography method of low cost that utilizes more than one form of interactions (hydrophobic, electrostatic, thiophilic interactions or hydrogen bond) between the stationary phase and solutes in the liquid phase, providing good selectivity and adsorption capacity. The aim of this work is to investigate the potential of ortho-phospho-L-tyrosine (OPT) as new ligand for purification of polyclonal IgG from human plasma. The contribution of carboxylate (negatively charged) and phosphate-aromatic (negatively charged-partially hydrophobic) groups in capturing of human IgG and the effect of the operating conditions (buffer system, pH, and ionic strength) on the capacity and the selectivity of the adsorbents will be studied. The development of the new adsorbent for purification process by MMC is relevant, constituting an important contribution with potential application in plasma fractionation and kits of depletion of IgG from human serum aiming proteomic studies.
