Prostatic lesions, inflammation and aging: effects of anti-inflammatory therapies in transgenic adenocarcinoma of the mouse prostate model and in senile FVB mice

Abstract

Senescence is related to changes in the hormonal environment causing to morphological and functional prostatic disturbance. The aim of this work was to evaluate and compare the effects of anti-inflammatory treatments, administering celocoxib and goniothalamin on the structural and molecular biology of the ventral prostate of senile mice (FVB) and TRAMP. The IL-6, IL1²- e IL-17, TNF-2, 4HNE, CD3, inducible nitric oxide synthase, ciclooxigenase 2, signal transducer and activator of transcription-3, nuclear factor kappa ²(NF-ºB), insulin-like growth factor-1 reactivities will be evaluated. 72 male mice FVB and 96 TRAMP were divided into groups: Young Control (YC): 24 animals (FVB)/ 8 weeks old will be subdivided and received carboxymethylcellulose 0.5% and PBS; Senile Control (SC): 24 animals (FVB)- 48 weeks old - the same treatment group YC. Senile Celecoxib (SNCEL): 12 animals (FVB )-48 weeks old mice- 10mg/Kg/day of Celecoxib; Senil Goniothalamin (SNGTN): 12 animals (FVB) - 48 weeks old mice -150mg/Kg/dia of Goniothalamin; TRAMP Control 1 (T1): 24 animals (TRAMP)- 8 weeks-old mice will receive the same treatment group YC; Tramp Control 2 (T2): 24 animals (TRAMP) -8 weeks old mice- will receive the same treatment group, and will be sacrificed at 22 weeks old. Tramp 1/ Celecoxib (T1CEL):12 animals (TRAMP) - 8 weeks old mice - the same treatment group SNCEL; Tramp 1/ Goniothalamin (T1GTN): 12 animals (TRAMP) -8 weeks old mice - the same treatment group SNGTN; TRAMP 2/Celecoxib (T2CEL) e TRAMP 2/ Goniothalamin (T2GTN): 24 animals (TRAMP), 8 weeks old mice - the same treatment group T1CEL e T1GTN, respectively, which will be sacrificed at 22 weeks old. After 30 days, the ventral prostate will be collected and analyzed by morphological, Immunohistochemical, Immunoenzymatic, Raman spectroscopy and Western Blotting analyses. (AU)
