Abstract
The effects of different concentrations of nitrous oxide (N2O) and oxygen on hematosis, bispectral index, cardiorespiratory and intracranial parameters in propofol-anesthetized pigs maintained in spontaneous breathing or in pressure-controlled ventilation with or without PEEP will be evaluated. For this, forty eight animals, males or females, were randomly divided into six groups: GN10 (FiO2 = 0.9 and N2O = 0.1), GA10 (FiO2 = 0.9 and compressed air = 0.1), GN50 (FiO2 = 0.5 and N2O = 0.5), GA50 (FiO2 = 0.5 and compressed air = 0.5), GN70 (FiO2 = 0.3 and N2O = 0.7), GA70 (FiO2 = 0.3 and compressed air = 0.7). For all pigs, azaperone (2.0 mg/kg) will be administered as premedication. Propofol will be used for induction and maintenance of anesthesia (0.4 mg/kg/min). After endotracheal intubation, the probe Magill will be coupled to inhalation anesthesia machine for the supply of gaseous mixtures at concentrations recommended for each group. After 125 minutes of anesthesia induction, rocuronium (0.6 mg/kg followed by 0.6 mg/kg/h) will be administered. Immediately, mechanical ventilation using pressure cycled (15 cmH2O) will be started. The variables observations will be conducted in three phases. In the first stage (M1), the parameters will be assessed 40 minutes after anesthetic induction (M10) and then at 15-minute intervals (M115 to M160). In the second phase (M2), the measurements will be performed 140 minutes after anesthesia induction (M20) and then at 15-minute intervals (M215 to M260). In last stage (M3), the analysis will be started 125 minutes after anesthesia induction (M30 = M160), 60 minutes after the beginning of controlled ventilation (M360 = M260) and then at 15-minute intervals (M375, M390 and M3115). After analysis of parameters at M360, PEEP of 5 cmH2O will be supplied. Numerical data will be subjected to two-way and one-way analysis of variance (ANOVA) followed by Bonferroni test (p<0.05). (AU)
