Polymorphims and expression of the immune system genes associated with mastitis in dairy buffaloes

Abstract

In Brazil, buffaloes stand out in dairy farming due to profit generated. However, economic losses due to changes in the production and composition of milk, making it less suitable for consumption and processing, are caused by mastitis, an infection of the mammary gland caused by a reaction to metabolic and physiological changes, trauma, or microorganism's invasion. Thus, the objective of this study is to evaluate in buffaloes with mastitis the gene expression level of genes lactoferrin, transferrin, ²-defensins and interleukin, and identify, polymorphisms in genes DRB2, DRB3, DMA and DMB. For the analysis of gene expression, samples of milk from animals diagnosed with mastitis and free of infection animals will be collected for RNA extraction, cDNA synthesis and validation expression profiling by qRT-PCR. The study of the expression of these genes related to active immune system in mammary gland can become an important tool for the developing of strategies aimed at preserving the health of the udder. For the study of polymorphisms, the samples of dander will be collected for DNA extraction, PCR and sequencing. MHC complex genes are related to the innate and adaptive system of the animal, so the identification of polymorphisms in these genes may provide relevant information about the allelic and genotypic variability of a given population, and provide markers for susceptibility / resistance to disease economically harmful to livestock, such as mastitis.
