Killer toxin production by Sporobolomyces koalae (ACBL-77) and Saccharomyces cerevisiae (ACB-K1) as mode of action against postharvest pathogens

Abstract

In order to understand as the killer toxins produced by Sporobolomyces koalae (ACBL-77) or Saccharomyces cerevisiae (ACB-K1) could kill sensitive yeast strain (S. cerevisiae NCYC 1006) including the pathogens Penicillium digitatum and Geotrichum citri- aurantii, the extracellular killer toxins in the supernatants of cell culture of yeast isolates will be purified and their antagonistic activities tested. The killer toxin purified from S. koalae will be characterized and their molecular mass will be estimated according to the data from SDS-PAGE. The optimal conditions for action of the purified killer toxin against the pathogens, as well as the identification of the gene encoding it will be evaluated. The identification of the type of killer toxin of S. cerevisiae (ACB-K1) will be accomplished by PCR amplification using specific primers of proteins known and produced by S. cerevisiae (K1, K2, and K28), and subsequently sequencing will be performed. If the amplification it is not possible, the characterization of killer toxin produced by ACB-K1 will be held following the same steps described for isolated ACBL-77. (AU)
