Ikaros: role in the BCR- signaling in B-1 cells and association with chronic lymphocytic leukemia

Abstract

Ikaros is considered an important regulator of lymphoid development and differentiation. Several reports reinforce the hypothesis that Ikaros expression is deregulated in human leukemias. It has been suggest the role of Ikaros in the regulation of the transcription of genes that are involved in the BCR signaling. The absence of Ikaros is in detrimental for activation of BCR signals. The Ikaros activity is regulated by kinases and phosphorilases, such as Btk. It is known that Btk deficiency reduced the number of B-1 cells. Additionally, some authors have demonstrated that BCR engagement in B-1 cells results in low proliferation rate and apoptosis induction, opposite to observed in B-2 lymphocytes. Interestingly, the absence of BCR signals diminish B-1 cell population, otherwise the over activation of this pathway is related to proliferative disorders. Despite of several reports demonstrated that some BCR signaling components are differentially expressed in B-1 and B-2 lymphocytes, a clear explanation about how this signaling pathway is regulated has not emerged yet. Considering this, herein we purpose to investigate the role of Ikaros in the regulation of BCR signaling, aiming to clarify some aspects about the participation of B-1 cells in the leukemia. (AU)
