Abstract
The presence of emerging contaminants in water bodies has been attracting the attention of international scientific community, mainly due to the lack of toxicological data available considering exposure to low doses. In this context, investigation of minor effects using early life stages of development in Zebrafish, specially High-throughput screening analysis have received special international recognition, considering the current discussions on the number of animals used in scientific research, because these analysis are able to evaluate multiple parameters simultaneously. The main objective of this project is the evaluation of damage on neurobehavioral and development of Zebrafish embryos and larvae exposed to psychotropic drugs, herbicides and dyes, after the implementation of High-throughput screening analysis in the laboratory of Environmental Toxicology, using a video surveillance software, DanioScope, requested in this proposal. In addition, we propose to complement the neurotoxicity studies using other video surveillance tool (Zebrabox), to monitoring larvae pattern of swimming, in collaboration with Prof. Dr. Amadeu Soares, University of Aveiro, Portugal. Finally, we also propose the evaluation of alterations on gene expression related to neurotoxicity in Zebrafish, which are also expressed in humans, as well as determination of the enzyme cholinesterase activity. Thus, this project aims to perform an integrated investigation of neurotoxicity induced by representatives of emerging water contaminants, offering subsidies for decision-making regarding the presence of these compounds in aquatic environments. Additionally, the results will be useful in establishing safety standards for these compounds, also considering human and aquatic life health. Moreover, the approval of this proposal will improve the research in the Environmental Toxicology Laboratory, as well as complement the tests already carried out. The project will also contribute greatly to the formation of specialized human resources, considering that masters and doctoral students, and post-doctoral students will participate. Finally, the project will encourage the collaboration between national and international researchers. (AU)
