Abstract

The mitochondrial unfolded protein response (UPRmt), it is recognized as an adaptive intracellular pathway in response to stress, which ensures the integrity and function of the mitochondrial proteome through the mitochondrial chaperone and protease action. Mutagenic studies have shown that the disturbance caused by the stoichiometric imbalance between the mitochondrial DNA proteins (mtDNA) and proteins of the electron transport chain encoded by nuclear DNA (nDNA), is able to activate the UPRmt. In organisms including C. elegans and Drosophila the mitonuclear imbalance results in the improvement of cellular metabolism, mitochondrial function and increased longevity. The pharmacological induction of mitonuclear imbalance activates the UPRmt leading to increased biosynthesis and mitochondrial function, improving the oxidative capacity in tissues such as the liver and muscle, improving the functional capacity of these organs. Although the induction of UPRmt mediated mitonuclear imbalance mechanism appears to be a well-conserved biological process and which progresses in improvement in bodily functions, the effects of physical exercise on UPRmt and the mitonuclear imbalance have not yet been explored. Thus the current project are aimed at evaluating the UPRmt markers and mitonuclear imbalance in the hypothalamus, heart, liver and skeletal muscle of trained mice, relating this phenomenon to the mitochondrial function in these tissues. The development of this project will contribute significantly to the understanding of the role of UPRmt on organic adaptations induced by physical training. (AU)
