Assessment of effects potential anti-adipogenic, antioxidants and anti-inflammatory of conjugated fatty acids in cell culture

Abstract

Currently, numerous studies have shown that the secondary metabolites of plants, in particular conjugated fatty acids (AGCs) to modulate pathways involved in lipid metabolism and obesity, having in common inflammation and oxidative stress. Our research group has been studying the composition and some biological effects of foods rich in AGCs, as the oils of seeds of pomegranate and bitter-gourd. The results are promising and indicate potential anti-oxidant and anti-inflammatory effects. This work aims to advance in the cellular level research under the action mechanisms referring to metabolism of the AGCs, as well as its anti-oxidant and anti-inflammatory activities. Initially will be used Caco-2 cell line to assess the transport and absorption these intestinal level, as well as assays that evaluate the antioxidant and anti-inflammatory activity thereof. Then these activities will be studied in hepatic cell lines (HepG2) and fat (3T3). Cell viability analysis, quantification of fat by Oil Red and free fatty acids, hormones leptin and adiponectin, reactive oxygen species and lipid peroxidation, antioxidant enzymes and inflammatory cytokines and their gene expression, besides the expression of genes involved in lipid metabolism will be performed. The results will contribute to the discussion on the effects of different isomers of AGCs in food as well as their possible mechanisms of action for the metabolism of fatty acids and their antioxidant and anti-inflammatory effects; that still present inconclusive results.
