Participation of the hippocampal formation in acquisition, consolidation, evocation and extinction on conditioned suppression: clinical correlations

Abstract

Previous studies from our group have demonstrated that the dorsal hippocampal formation (DHF) has been one of the main targets for modulation of drugs during acquisition and extinction of conditioned suppression, a suitable animal model for investigating the behavioral changes and pharmacological mechanisms that underlie fear memory and anxiety. The participation of DHF in these process, will be evalueted by: (i) temporary inactivation of CA1, for infusion of muscimol, 30 min before the acquisition (training) or recall (test) of conditioned fear; ii) differential expression of proteins involved in different signaling pathways in the DHF of rats after acquisition, consolidation and recall the fear conditioning and acquisition, consolidation and recall of fear extinction by Western blot and immunohistochemistry techniques; iii) analysis of the participation of DHF in systemic consolidation of fear memory through the analysis of protein profile by proteomics of the DHF of rats submitted to retention test 48 hours or 40 days after the conditioning fear. This analysis will assess the hippocampal dependency of memory recall. In addition to providing our understanding about memory ansd anxiety mechanisms, this study will contribute substantially to the strengthening of research in our laboratory and therefore, of the graduate programs, because it proposes the inclusion of new methods of analisys and interaction with differents laboratories. Furthrermore, it will enable the consolidation of the institutional and pedagogical projects strengthening the performance of undergraduate students and graduate students in the laboratory. (AU)
