Abstract The aim of the current study is to evaluate, in vitro and in situ, the effect of the Er:YAG laser parameters (frequency,water flow rate and energy) on enamel changes after cariogenic challenge. For in vitro study, sound human third molars will be cut for selection of 375 specimens, which will be randomly assigned into twenty five groups (n=15), being 24 experimental groups and one control. After cavity preparation, the samples will be filled with composite resin and submitted to cariogenic challenge. Subsequently, they will be cut for microhardness and light polarized microscopy analyses. For in situ study, 60 human enamel and 60 bovine enamel samples will be randomly assigned into fifteen volunteers. The samples will be allocated into four groups according to cavity preparation. The three experimental parameters that demonstrate higher acid resistance for in vitro study will be employed for cavity preparation for in situ study. The control group will be prepared with diamond bur in a high-speed handpiece. The specimens will be filled and put on palatal device which will be inserted in the oral cavity of the volunteers for cariogenic challenge. After cariogenic challenge, the samples will be sectioned for microhardness and light polarized microscopy analyses. The obtained data will be analyzed for choose the statistical method for analysis.
