Effect of demineralization time, remineralization treatments and caries challenging on enamel caries-like lesions produced in vitro

Abstract

This Project will be developed in 3 independently steps. The mains objectives will be to identify the demineralization time needed to produce enamel caries-like lesions on bovine enamel surfaces, using a demineralizing solution 50% hydroxyapatite satured, whose clinical characteristics will be similar to those found 1, 2 and 3 codes on ICDAS II (2009); to evaluate the effect of 4 different remineralization procedures (artificial saliva- Control; Fluoride (5% NaF varnish-Duraphat, Colgate-Palmolive, S. Bernardo do Campo, SP, Brasil; MI-GC toothpaste with CCP-ACP (Recaldent®); MI Plus-GC, with CCP-ACP (Recaldent®)+Fluoride on caries-like lesions; and, to evaluate the effect of ICDAS codes (1,2,3) for primary caries produced in vitro and remineralized by 4 different remineralization procedures and then, submitted to cariogenic challenge using a pH cycling model. It will be used the following evaluation parameters in order to identify the enamel caries-like lesions production, remineralization and cariogenic challenge effects: Clinical parameters: visual inspection - ICDAS II (2009) and X-ray; Morphologic Parameters: surface and depth of lesions -SEM, Polarized Light Microscopy, m-CT, Confocal Laser Scanning Microscopy; Chemical Parameters: m-EDX; and Cross-sectional Transversal Microhardness (KHN). These parameters will be statistically compared using appropriated statistical tests in order to verify the association between them. (AU)
