Abstract

While it has been clearly demonstrated the role of functional genomics on tumorigenesis, the biological behavior of some oncogenes, tumoral invasiveness, DNA repair kinetics and content along time course oral cancer progression are not fully established. Thus far, the goal of this project will be to investigate the role of some oncogenes, cellular proliferation control, DNA repair kinetics and content during experimental rat oral carcinogenesis. In this regard, a total of 30 male Wistar rats will be exposed to carcinogen 4-nitroquinoline 1-oxide by drinking water at 50 ppm dose and then, they will be sacrificed at four, 12 and 20 weeks post-treatment. A total of 30 rats will be used as negative control. Herein, it will be evaluated mutations in H-ras and K-ras oncogenes, expression of p21RAS, ki-67 e alpha-SMA, through immunohistochemistry and global genome content and DNA repair system performance by single cell gel (comet) assay during rat tongue carcinogenesis.
