Abstract

The majority of medicines administered by the oral route is in the solid form and, among them, tablets represent 80% of the world market. Controlled drug delivery systems (CDDS), specially the solid forms obtained from hydrophilic matrices, play important role due to the possibility of incorporation of great amounts of drug, economy and easy of processing and allowing reproducible drug release profiles. The properties of hydrophylic polymers can be altered by cross-linking, originating hydrogels, which are swellable in aqueous media, making possible the effective control of drug release. Recently, in our laboratories, high amylose was cross-linked by reaction with sodium trimetaphosphate (TMPS), originating non-compacted solid systems, which prolonged dichlofenac release up to 24 h. Pectin can also be cross-linked with the same reagent and, inclusive, in mixtures with high amylose. However, such excipient has not yet been used for the preparation of CDDS. Thus, the objective of this project is to cross-link the referred polymers, isolated and in mixtures and extensively characterize the products, with which matrix tablets containing nimesulide, a class II model drug (low solubility and high permeability) will be prepared. The tablets will be characterized in relation to physical properties and "in vitro" drug release, which mechanisms will be analyzed by mathematical models.
