Effect of vicilin isolated from the species Glycine max, Vigna unguiculata and V. angularis in lipid metabolism of rats

Abstract

Many studies have shown proteins as a biologically active agent in addition to their known nutritional role. In turn, the proteins of some legumes have received special attention due to their involvement in metabolism and the prevention of lipid disorders, especially hypercholesterolemia, hypertriglyceridemia and hyperglycemia. Different approaches have shown an association of the 7S protein fraction (vicilin) of soybean to certain alterations in lipid metabolism in animals and humans. Furthermore, some similarities between vicilins of different species such as composition characteristics, amino acid sequence and other properties have been registered. Accordingly, this project proposes to investigate the action of proteins of vicilin type, isolated and purified from species of Glycine max., Vigna angularis and Vigna unguiculata in the lipid metabolism of animals subjected to a hypercholesterolemic diet. Wistar male rats will be divided into 9 experimental groups (n = 9). A control group (STD) will receive a standard diet, a second group will be submitted to (HC) a hypercholesterolemic diet of reference, and the other groups will receive HC diet followed by daily doses of the isolated proteins that will be administered by gavage. The last group will be treated with 50mg/kg body weight/day of a hypolipidemic drug. For all groups parameters such as growth, food consumption, faecal excretion and deposition of fat will be examined. Triglycerides, total cholesterol and lipoprotein fractions in the plasma and liver will be quantified, and associated to lipid metabolism the activity of the enzyme HMG-CoA, analysis of gene expression of receptors (SRBP-1, PPAR± and the LDL receptor) and enzymes in the liver (fatty acid synthase, 7-± hydroxylase, HMG-CoA reductase and malic). It is hoped that the study will contribute to a greater understanding of the possible effects and their mechanisms of action and can lead to aggregation of benefits for issues of public health and economic-sector social.
