Abstract

Introduction: The diet plays an important role in cardiovascular disease and some foods, such as amaranth and cowpea, have proven ability to reduce serum cholesterol. This effect is related to peptides formed during digestion of their protein. Bioavailability and the fate of these peptides after digestion are clearly complex, and their target tissues are unknown. Objectives: To assess the bioavailability of peptides from amaranth and cowpea and verify their relationship to the absorption and metabolism of cholesterol. Methodology: Synthetic peptides will be used to verify gene expression related to cholesterol metabolism in Hep G2 cells. Besides, in vivo experiments will be conducted. In these experiments, animals will be fed protein isolated from the cowpea and from amaranth, and the concentration of target peptides will be monitored in the blood. Furthermore, there will be checked several metabolic aspects and gene expression of various receptors, transporters and enzymes in various tissues of animals fed hypercholesterolemic diets containing amaranth or cowpea proteins as unique feed protein sources. There will be determined in the plasma: cholesterol and fractions; in the faeces: cholesterol and bile acids; in the liver: cholesterol, total lipids, HMGR activity, and quantification of protein expression of HMGR, SREBP-2, NPC1L1, ABCCG5, ABCCG8, ABCA1, ABCG1, LDL receptors, PEPT1 receptors; and in the gut: protein expression and quantification of NPC1L1, ABCCG5, ABCCG8, ABCA1, ABCG1 and PEPT1 receptors.
