Abstract

Viral replication involves virus entry in plant cell and hijacking the host machinery for necessary protein production to give rise new viral particles. Molecular studies of plant viruses have been made, for example, RNAs sequencing, subgenomic RNA detection and determination of strategy for expression of the virus genome. However, several aspects related to the function and structure of some proteins, as well as its relation to the viral replication cycle, are still unknown. Thus, the aim of this study is a high-scale production and a high-purity purification of viral proteins in its native state. The proteins will be expressed in Escherichia coli, aiming the high-resolution determination of its three-dimensional structures, providing relevant informations about their roles and functions. The following proteins will be studied: Cole latent virus capsidial protein (CP), Pepper ringspot virus RNA-dependent RNA polymerase (RdRP) and Pepper ringspot movement protein (MP).
