Abstract

Abdominal pain may be consequent to several causes, among the various possibilities need to be attentive to the intestinal ischemia. In the digestive tract ischemia / reperfusion (I/R-i) entails morphological changes in enteric neurons. This work aims to analyze the different classes of neuronal and P2X7 receptor in the enteric nervous system of the rats ileum subjected to I/R-i. Intestinal ischemia is obtained by the obstruction of blood flow ileal arteries within 60 minutes followed by reperfusion periods of 4, 24 hours, 7, 14 and 28 days. In the sham group there is no occlusion of the ileal arteries. The tissues will be prepared by immunohistochemical methods for double and triple marks of P2X7 receptor with Nitric Oxide Synthase (NOS), calbindin (Calb), acetylcholine transferase (ChAT), calretinin (Calr), Neuropeptide Y (NPY) and Vasoactive Intestinal Peptide (VIP) and P2X7 receptor, NOS, of ChAT, the Calr, NPY and VIP with anti-pan-neuronal anti-HuC/D. The qualitative and quantitative analysis from of double or triple labelling, the neuronal density and area of neuronal profiles will be obtained from the fluorescence microscopes and/or confocal scanning laser. Also, be made the analysis of protein expression of the P2X7 receptor in different periods of reperfusion, analysis of apoptosis/necrosis analysis of myeloperoxidase, ultrastructural analysis of organelles and analysis of enteric plasticity.
