Abstract

Melanoma, the most fatal skin cancer whose incidence has increased rapidly, develops from malignant transformation in melanocytes. In virtue of its low response to treatments traditionally adopted as chemotherapy and radiotherapy, the development of new therapeutic strategies is of particular importance. Although studies have shown promising anti-tumor effects of synthetic phosphoethanolamine (PEA), the mechanisms by which they occur are presently unknown. The aim of this study is to assess the activity of synthetic PEA in experimental murine melanoma (B16F10 cells implanted in C57BL/6 mice). The animals will be treated orally with PEA (2.5, 5 and 10mg/ml) for 20 consecutive days or with PBS only (negative control). At the end of treatment, animals will be sacrificed, the dorsal tumors analyzed, the macroscopic internal lesions (metastases) identified, measured and photographed. Tumor samples from different experimental groups will be processed for immunohistochemical analysis, through which proliferation and death of tumor cells, and the presence of tumor-infiltrating immune cells (macrophages and lymphocytes) will be studied. Cells will be isolated from popliteal lymph node and/or spleen and after stimulation in vitro, pro-or anti-inflammatory cytokine levels in the supernatants of these cells will be measured. The blood will also be collected and used for hematological analysis. (AU)
