STUDY OF THE CYTOTOXICITY OF THE DODAB LIPOSOMAL FORMULATION WITH THE COMPOUND SYNTHETIC PHOSPHOETHANOLAMINE IN TUMOR CELL LINES OF HEPATOCELLULAR CARCINOMA

Abstract

Tumor cells are metabolically adapted for rapid growth and proliferation under conditions of low pH and oxygen tension in which normal cells would grow only poorly or could not survive. Some alternatives of treatment for these cellular dysfunctions has been applied as repair method, for example, mitochondrial system in tumor cells may be dramatically improved, just changing the availability of energy substrates. Our research group has been using the synthetic phosphoethanolamine (PHO-S), as adjuvant treatment for cancer in vitro and in vivo experimental models. Despite the proven efficacy using the PHO-S, in murine and human melanoma cell lines, and breast adenocarcinoma, the antitumor treatment could be maximized with the use of liposomes, and other carriers based in drug delivery systems with the aim of achieving tumor cells with greater efficiency and specificity. Thus, this project aims to evaluate "in vitro" cytotoxic effect of the DODAB liposomal formulation with the compound PHO-S in hepatocellular carcinoma cell lines aiming at future utilization this model in the treatment of hepatocellular carcinoma, as well as, check whether there is synergism between liposomal formulation on the inhibition of cell growth. Two hepatocellular carcinoma cell lines will be cultivated (hepa1c1c7 e HepG2) and four normal cell lines (fibroblasts, hepatocytes, osteoblasts and leukocytes), then will be treated with the DODAB liposomal formulation with the compound PHO-S, with different concentrations and treatment times. Then, assessments will be conducted to determination of the inhibitory concentration (IC50%) this formulation on tumor and normal cell lines. That way, the execution this project will provide relevant information for maximize development of specific technologies to the induction of cytotoxicity to tumor cells.
