Abstract

Breast cancer is the second most deadly cancer for women worldwide. Currently, conventional approaches to the treatment of breast cancer include radiotherapy, chemotherapy and hormone therapy. The resistance of tumor cells to chemotherapy is a major obstacle to the clinical success of cancer therapy. Synthetic phosphoethanolamine is a phosphorylated molecule artificially, which makes it different from its current level of absorption by approximately 90%, with anti-inflammatory properties and apoptotic molecules. Our group has developed several experiments involving the Pho-S with the liposomal vesicles and the results are fully satisfactory as to the efficacy of liposomal formulation. In this project the aim is to evaluate the anti-proliferative and pro-apoptotic effects in vitro liposomal formulation DODAC with synthetic phosphoethanolamine in human breast tumor cell line, MCF-7 effects. The effects of phosphoethanolamine cytotoxicity in tumor cell lines will be evaluated for viability by the MTT colorimetric assay and detection of IC50% by free radical production (LPO), changes in cell death pathways by flow cytometry. The intraestruturais changes by scanning electron and confocal laser microscopy. Marking of lysosomal vesicles and mitochondrial electrical potential probe rhodamine-123. The arrangement of the cytoskeleton by staining with phalloidin and quantification of cells in the senescence ²-galactosidase staining. (AU)
