Immediate reconstruction of peri-implant defects in implants after tooth extraction. histomorphometric analysis in dogs

Abstract

The aim of this study will be to evaluate the regeneration at peri-implant defects using different biomaterials in conjunction or without BMP-2 (osteomorphogenetic protein). For this study, the mandible of 8 Labrador dogs will be used, and implants will be installed immediately after tooth extraction in the distal alveolus of the second, third and fourth premolars and of the first molar, bilaterally. After implant installation, standardized buccal defects will be created and filled with different biomaterial. The following 8 groups will be obtained: 1) COA: clot (control site); 2) AO: autologous bone; 3) BCP: biphasic calcium phosphate (mixture of 60% hydroxyapatite and 40% of beta-tricalcium phosphate; Bone Ceramic®); 4) BCP-BMP: biphasic calcium phosphate + BMP-2; 5) EN: deproteinized bovine bone mineral (Endobon®); 6) EN-BMP: Endobon® + BMP2; 7) BO: deproteinized bovine bone mineral (Bio-Oss®); 8) BO-BMP: Bio-Oss + BMP2. Immediately after implant installation as well as at the end of the experiment (3 months), standardized periapical x-rays of each implant area will be taken to evaluate bone regeneration. After 3 months postoperatively, the animals will be sacrificed and perfused through the carotid arteries with fixative. The mandible will be removed and individual bone blocks containing the implants and the surrounding soft tissue will be obtained, processed for histology and analyzed. Bone regeneration at the buccal aspect of the implants and morphometry of area will be evaluated and tested with ANOVA.
