ANALYSIS OF THE PERI-IMPLANT BONE TISSUE OF IMPLANTS PLACED IN THE REPARATIVE TOOTH SOCKET OF RATS TREATED WITH ZOLENDRONIC ACID

Abstract

Bisphosphonates (BPs) are anti-resorptive drugs prescribed for individuals with bone disorders that affect bone remodeling, however the dose and prolonged use of these drugs can lead the development of medication-related osteonecrosis of the jaws (MRONJ). The aim of this project will be to evaluate the peri-implant bone tissue of implants placed in reparational alveolar bone after local preventive therapies during treatment with zoledronic acid. For this purpose, 96 male Wistar rats will be treated with 4 applications of 0.035mg/kg of zolendronic acid (ZOE) through the caudal vein with 15 days intervals between applications and subsequently their mandibular left first molars were extracted. After the extraction, animals will continue with treatment with ZOE applications, as well as after implant placement until euthanasia. The animals will be divided into six groups (n=8) according to the treatment of the dental socket: GC (Control - will not be applied ZOE, tooth socket filled with blood clot), GZ (blood clot), G² (²-tricalcium phosphate), G²D (²-tricalcium phosphate + 10% doxycycline gel), G²DP (²-tricalcium phosphate + 10% doxycycline gel + antimicrobial photodynamic therapy) and GDP (10% doxycycline gel + antimicrobial photodynamic therapy). After 28 days of extractions, each animal will receive an implant (2.2mmx4mm) in the region of reparational alveolar bone of the mandibular left first molars, totaling 96 implants installed. After 28 days of installation of the implants, 48 animals will be anesthetized and in vivo biomechanical analysis will be performed by means of removal torque, followed by euthanasia. And the implants removed by means of removal torque will be submitted to topographic characterization by means of scanning electron microscopy, coupled to the X-ray dispersive energy spectrometry system (SEM-EDX) for qualitative and semiquantitative analysis of tissue neoformed bone with the measurement of the elements calcium (Ca), phosphorus (P) and oxygen (O). And the other 48 animals belonging to the mineralized section methodology will be used confocal laser microscopy with applications of fluorochromes (calcein and alizarin red). All values obtained will be tabulated and submitted to statistical analysis, considering p<0.05.
