Abstract

With this objective is to evaluate the expression of inducible nitric oxide synthase (iNOS) and metalloproteinase-9 (MMP-9) in rabbit corneas ulcerated with alkali and treated locally with ketorolac tromethamine. After general anesthesia, will be produced ulcers with sodium hydroxide in twelve rabbits. The left eye from each animal is treated with Ketorolac tromethamine 0.5% and the right eye with saline solution 0.9%, at regular intervals of 6 hours. Six animals corneas are harvested after 24 hours of initial lesions, the rest, the same will occur after reepithelialization of them. Corneas are removed, formalized and embedded in paraffin. Polyclonal antibodies are employed for immunohistochemical staining for the identification of the expression of iNOS and MMP-9, by the technique of streptavidin biotin peroxidase complex, and revealed by diaminobenzidine. The immunostaining score is assessed semi-quantitative scale as absent (0), mild (1), moderate (3) and intense (4), for the presence of cells positive for iNOS and MMP-9. It will employ analysis of variance for repeated measures, followed by post-hoc evaluation and appropriate test according to the normality of the data. Possible correlations between the score of immunostaining of iNOS and MMP-9 are tested for Pearson. In all analyzes will adopt minimum significance level of p less than 0.05. (AU)
