Advanced Glycation products and DNA adducts related to oxidative stress and inflammation in a model of diabetic rats

Abstract

Diabetes mellitus has been considered globally one of the most challenging health problems in the 21st century, and the chronic complications of the disease are responsible for this situation. It is known that diabetes promotes oxidative stress, inflammation and the formation of advanced non enzymatic glycation end products (AGEs), resulting in damage to several biomolecules. There is a continuing interest in the development of more sensitive methods for the quantification of AGEs, injuries due oxidative stress and inflammation in diabetes, searching for new biomarkers to better understand the risks associated to this disease and for treatment strategies. To better understand the profile of DNA lesions that can occur in diabetics pacients, it will be analyzed in this project the formation of DNA adducts in a pathological condition of diabetes. So the adducts N2-carboxietil-2'-desoxiguanosina (CEdG); 1,N2-eteno-2'-desoxiguanosina (1,N2-[dGuo); 1,N6-eteno-2'-desoxiadenosina (1,N6-[dAdo); 8-oxo-2'-desoxiguanosina (8-oxodG); 8-nitro-2'-desoxiguanosina (8-NO2-dG); and 8-cloro-2'-desoxiguanosina (8-Cl-dG), related to oxidative stress, inflammation and AGEs processes will be quantified in tissues of diabetic rats, diabetic rats treated with insulin and non-diabetic rats for comparison. The study will provide a broader view of diabetes-related injuries in the body, comparing the results with the non pathological situation or treated diabetes, also enabling strategies for anti-AGE drugs tests.
