Immunohistochemical analysis of VEGF protein and microvessel density in the periodontium of diabetic rats treated with green tea

Abstract

There are several reports that investigated the relationship between Diabetes mellitus (DM) to the severity of periodontitis. These investigations showed the contribution of DM to the severity of periodontitis via its effects on vasculature, inflammatory and immune response, alteration in collagen synthesis, and genetic predisposition. The increased concentration of vascular endothelial growth factor (VEGF) has been considered an important factor in the pathogenesis of chronic and aggressive periodontitis. Multiple lines of evidence suggested that Green tea consumption is associated with a reduced risk of several human malignancies such as cancer and diabetes due to antiangiogenic effects. The green tea polyphenols with the epigallocatechin may exert inhibition of the activation of the VEGF/VEGFR axis by suppressing the expression of HIF and other growth factors. This study investigated the de relationship between VEGF, inflammation, and periodontal disease in DM and the possible therapeutic effects of green tea in the modulation of angiogenesis. Materials and methods: serial sections (4µm thickness) will be obtained from the hemi-mandibles of streptozotocin-induced diabetic rats that ingested or did not green tea (Camellia Sinensis) daily for 15, 30, 60, and 90 days (n=5 animals/group). In the mesial root of the first molar stained with hematoxylin-eosin will be evaluated the inflammatory process and alveolar bone loss. In the tissue sections obtained on silanized slides immunostained against VEGF and CD34 are evaluated, respectively, the percentage of VEGF-positive cells per mm ² of periodontal tissue and microvessel density (MD). Numerical data will be compared between groups using analysis of variance (ANOVA) and Tukey's post hoc test at a significance level of 5%. The Pearson correlation coefficient will be carried out to examine the degree relationship as well as direction sign (positive or negative) between the inflammatory infiltrate and alveolar bone loss variables versus the VEGF expression and MD.(AU)
