Abstract

The drug targeting to a specific organ represents a major challenge in the research and development of new controlled release systems. The use of bioadhesive polymers in form of multiparticulate systems, such as microcapsules, as well as its chemical modification by cross-linking, can turns into valuable strategies to improve the performance of colon-specific drug delivery systems. In this work, mixtures of gellan and pectin will be used to produce microcapsules containing ketoprofen as model drug, by ionotropic cross-linking with Al3+. Physico-chemical characterization of microcapsules will include analyses of encapsulation efficiency, size, shape and surface morphology. Thermal analysis will be used to assess possible interactions drug-polymer, as well as to determine the form in which the drug is encapsulated. The performance of microcapsules as drug release systems will be determined by in vitro release studies using media with pH values corresponding to different portions of TGI, followed by the determination of its release mechanism through appropriate mathematical models. The influence of variables: proportion of polymers mixture, polymer concentration and cation (Al3 +) in the experimental data obtained will be statistically evaluated.
