Abstract

In 2008, data from WHO showed that 1.5 billion of people were overweight. Recent research conducted by Ministry of Health revealed that 49% of Brazilians are overweight. Therefore, obesity became a big problem of public health. There are evidence that maternal food consumption can cause impairment in adult life of the offspring, like a high risk of developing obesity and metabolic alterations. Furthermore, mother's high fat diet seems to be able of making changes in methylation and expression of opioid-related genes of descendents. However, few studies have been investigating the influence of maternal high fat diet on maternal behavior and perinatal development of the young. The influence of a high fat diet on opioid system is poorly understood too. This study aims to investigate the influence of high fat diet in the reproduction context, focusing on mother-pups interaction and its immediate and late effects. For this, female rats will be divided randomly in to groups: HF group that will be feed with high fat diet (45% of fat) from day 0 of pregnancy to 21th day of lactation (PND21); and control group that will receive a pattern diet. In PDN21, each litter will be subdivided in 2 groups: one that will continue receiving the same diet of the mother, and another one that will receive a different diet of the mother resulting in 4 different experimental groups: (C-C control-control; C-HF control-high fat; HF-C high fat; HF-HF high fat-high fat). It will be evaluated the glucose levels of the mother, and the glucose and insulin tolerance tests will be realized with her. In PND5 and 10, it will be evaluated the maternal behavior with and without previous manipulation. In PND21, mothers will be sacrificed and their brain will be removed for analyses of the expression of opioid receptors genes in hypothalamus. The physical development of the offspring will be evaluated, and the maternal behavior of female will be analyzed in adult life.
