Evaluation of plasma concentrations of TNF-alpha and il-6 in ovariectomized rats chronically treated with sodium fluoride

Abstract

In the last few years there has been a significant reduction in dental caries rates in several regions of the world, a fact that has been attributed to the use of fluoridated products, such as toothpaste. Simultaneously, there was an increase in the prevalence of dental fluorosis. Study performed in 25 patients (15 to 30 years old) with endemic fluorosis showed that 40% of these had impaired glucose tolerance, however, this abnormality was reversed by removal of excess fluoride in the drinking water. The NaF causes inhibition of glycolysis, reduced insulin secretion and hyperglycemia. Many of these responses suggest that NaF can cause insulin resistance. If this is confirmed, can be recommended decrease the fluoride concentration in toothpaste used mainly by children with diabetes, because the ingestion of fluoridated toothpaste can lead to worsening in health condition of these children. It is known that fluoride may alter the metabolism of carbohydrates, it is fundamental evaluate the effect of the chronic fluoride intake on 1) the IRS-1 serine phosphorylation status in liver; 2) plasma concentration of cholesterol and triglycerides. For this, male Wistar rats (1 month old) castrated were used. Thirty days after castration, the animals will be randomly divided into two groups: 1) Control group (CN), that will be subjected to treatment without NaF, but with a NaCl solution (9.54 mg/kg bw) containing the same amount of sodium in relation to the group NaF, 2) NaF group (FN), that will be submitted to treatment with NaF solution (4.0 mg F/kg bw) in the drinking water and feed for 42 days. After 6 weeks, the experiments will be conducted to quantify the IRS-1 serine phosphorylation status, after insulin stimulation, in liver tissue by Western blotting method and to evaluate the plasma concentration of cholesterol and triglycerides by ELISA method.
