Abstract

Antithrombin is a plasma glycoprotein belonging to the serpin family that plays a key role in controlling hemostasis, acting as an important inhibitor of blood coagulation by inactivating thrombin and other serine proteases, such as factors VIIa, IXa, Xa, XIa and XIIa. In addition to participating as a major physiological inhibitor of blood coagulation, antithrombin has anti-inflammatory activity, which is independent of its anticoagulant action. This serpin plays its anti-inflammatory role by reducing the synthesis of proinflammatory mediators and modulation of leukocyte activation and its interaction with the vascular wall. Our group has isolated and characterized the plasma antithrombin of Bothropoides jararaca snake and the results showed that this inhibitor is similar to antithrombins described in other species due to its biochemical and functional characteristics. This molecule also has anti-inflammatory activity and is able to inhibit the formation of paw edema and migration of cells to the peritoneal cavity induced by carrageenan in mice. In order to deepen the studies on the antithrombin of this snake, the aim of this work is to conduct a comparative proteomic analysis between the peritoneal exudate of mice treated with this molecule and mice that received no treatment, in order to identify its mechanism of inhibition and the mediators involved in this process.
