Abstract

Metastases are the major cause of death among cancer patients, especially due to therapies for these patients are extremely limited. By identifying the genes that control metastasis, it will be possible to predict the patients whose disease is likely to spread. Furthermore, once the genes that are critical to metastasis of breast cancer are known, an opportunity exists to develop therapies that target these genes and thereby provide these patients with more effective treatments. The aim of this work is to identify genes whose reduction leads to metastasis to secondary sites through a shRNA library screening. The poorly metastatic mammary tumor line 66cl4 will be infected with a lentiviral mouse shRNA library and the successfully infected cells will be inoculated into Balb/c mice. The metastatic nodules in these mice will be analyzed to identify the gene that has been knocked down. Once identified, in vivo and in vitro experiments will be conducted to validate the targeted gene as a metastasis suppressor gene and also elucidating the gene's function, respectively. The expression of the gene in human breast cancer will also be assessed to determine if it is a prognostic factor for outcome in breast cancer patients. (AU)
