Abstract

The chlorophyll retention has affected the production of soybean seeds in Brazil, mainly in the cerrado biome. As a result, has reduced the quality of lots, evidenced by decrease in germination and vigor, with losses in the establishment of appropriate population of plants at planting. Besides affecting the physiological quality of seeds, the presence of chlorophyll also exerts a negative effect on the quality of oil extracted. The process of chlorophyll degradation occurs naturally at the end stage of seed development. But this degradation is affected by ripening stage, by drying conditions, application of chemical desiccants and by climatic conditions, especially during periods of reserve accumulation and seed maturation. Despite the importance of the disappearance of chlorophyll in the seed, knowledge concerning the mechanism of degradation of this pigment is scarce. Identification of genes differentially expressed in soybean seeds that presents chlorophyll retention can be used in breeding strategies and biotechnology, and it is desirable to advance the understanding of this phenomenon, allowing the obtention of cultivars more tolerant to the occurrence of green seed in stress conditions. Thus, the objective of this phase of the study will be associate the presence of chlorophyll in soybean seed with genes differentially expressed in conditions of water stress associated with heat stress. The research project is part of a PhD project (2010/19878-8) linked to a project to assist the regular research (2010/20263-8) and will be conducted in the seed laboratory at the Wageningen University - Netherlands. The microarrays hybridized in Brazil will be analyzed and interpreted with the aid of bioinformatics tools and knowledge of biostatistics. Genes differentially expressed and identified with the use of microarrays will be selected and validated by real time PCR. These data obtained at Wageningen University will provide support for writing the doctoral thesis to be completed in Brazil. (AU)
